異硫氰酸熒光素可以使細胞內的蛋白質(zhì)著(zhù)色進(jìn)而把細胞染色。
異硫氰酸熒光素是生化試劑,也是醫學(xué)診斷藥物,主要用于熒光抗體技術(shù)中的熒光染料,能和各種抗體蛋白結合,結合后的抗體不喪失與一定抗原結合的特異性,并在堿性溶液中具有強烈的綠色熒光。用于醫學(xué),農學(xué)和畜牧等方面,可對由地細菌病毒和寄生蟲(chóng)等所致疾病進(jìn)行快速診斷。
異硫氰酸熒光素(fluorescein isothiocyanate, FITC) FITC純品為**或橙**結晶粉末,易溶于水和酒精溶劑。有兩種異構體,其中異構體Ⅰ型在效率、穩定性與蛋白質(zhì)結合力等方面都更優(yōu)良。FITC分子量為389.4,最大吸收光波長(cháng)為490~495nm,最大發(fā)射光波長(cháng)為520~530nm,呈現明亮的黃綠色熒光。FITC在冷暗干燥處可保存多年,是目前應用最廣泛的熒光素。其主要優(yōu)點(diǎn)是人眼對黃綠色較為敏感,通常切片標本中的綠色熒光少于紅色。
